No other lacZ mutant showed a marked difference in mutation rate between fpg and fpg backgrounds (that is, rates were comparable between HS101 and CSH1191, HS102 and CSH1192, etc.). The number of reversion mutants was much higher in HS1194 than in any other fpg strain. The inclusion of the pTRC99a p

  2022-03-11

To further investigate the molecular mechanisms that underlie glycine's β-cell protective effects through its antioxidative activity in diabetes, we used a H2O2/high glucose-induced apoptosis cell model. Similar effects on ROS generation of high glucose and H2O2 support the in vivo results, suggesti

• The dopamine substrate currents IDA IControl were determined

  2022-03-11

The dopamine substrate currents (IDA – IControl) were determined at different holding potentials (IV curves) in transfected HT22 cells (Fig. 5A). The plot for HT22 cells transfected with DAT exhibited the typical behavior of dopamine transporter currents (Ingram et al., 2002; Sonders et al., 1997),

• Hippocampal lesioned mice were less able to withhold respond

  2022-03-11

Hippocampal-lesioned mice were less able to withhold responding during a DRL-10 requirement (i.e. when the animals had to wait 10s between lever presses), and were thus less efficient than sham operated controls. In contrast, GluR-A−/− mice did not differ from wild-type mice when the DRL requirement

  2022-03-10

Integrin engagement triggers activation of several signaling pathway including Fak, which is recognized as the key signal transduction factor in the integrin signaling pathway that functions by activating downstream signaling in focal adhesion plaques. Whether changes in integrin-FAK signaling pathw

  2022-03-10

Histone modification is one of the major epigenetic mechanisms.8, 10 Specific histone modifications confer active or repressive transcriptional states. For instance, the trimethylation of Histone3-Lysine27 (H3K27me3) is a marker of tightly packaged heterochromatin and is associated with gene repress

Acknowledgments I thank Takeshi Sakaba for critical reading of the manuscript and helpful comments. This work was supported by JSPS/MEXT KAKENHI Grant Numbers 17K19466, 17H03548. Introduction The oral route of drug application is the main route in pharmacology since it is easy to handle and o

  2022-03-10

Here, we show that Jmjd3, a zebrafish JmjC domain-containing protein, possesses H3K27 histone demethylase activity in vivo, and down-regulation of jmjd3 leads to significant reduction of primitive and definitive myelopoiesis. Importantly, we have demonstrated that jmjd3 directly upregulates spi1 exp

  2022-03-10

Mammalian chromosomal DNA binds to various proteins and the DNA-protein complexes form DNA packaging units, called nucleosomes. Histones are the chief protein components of nucleosomes and perform pivotal functions in chromosomal gene regulation. Moreover, many types of chemical modifications of h

  2022-03-10

MK-3134 (3-[4-(1-cyclobutylpiperidin-4-yl)oxyphenyl]-2-methyl-5-(trifluoromethyl)quinazolin-4-one) was synthesized by Merck, as the structurally constrained analogue of MK-0249 (Nagase et al., 2008). This H3R inverse agonist has MW 457.49, seven HBA, and MLogP 4.58, with one violation in terms of dr

  2022-03-10

The glycolytic activator 6-phosphofructo-2-kinase/fructose 2,6-bisphosphatase 3 (PFKFB3) is well-known as a downstream substrate of hypoxia-inducible factor 1α (HIF-1α) signaling pathway [5,6]. Tumor hypoxia has long been associated with increased malignancy, poor prognosis and drug resistance. HIF-

  2022-03-10

In the adult normal spinal cord, the resident cells mainly include neurons, astrocytes, oligodendroglia and microglia (Lecca et al., 2012, Miyagi et al., 2012). After SCI, the lesion microenvironment is complex, which includes the functional changes of CNS resident cells and the infiltration of leuk

Recently, some published studies showed that HBV can induce autophagy in vitro and vivo [[19], [20], [21]]. The critical role of autophagy in HBV normal life cycle, such as HBV envelopment, has been gradually accepted by researchers from different laboratories [22,23]. HBV can trigger the autophagic

  2022-03-09

Because HIV does not infect neurons, the neurotoxicity in HAND is thought to result from the neuroinflammation evoked by the release of viral proteins and cytokines from infected microglia and macrophages (Ellis et al., 2007; Saylor et al., 2016). Viral tropism is determined by gp120 binding to CD4
